Maria Eleonora was born in 1599 in Königsberg (now Kaliningrad), the daughter of Elector of Brandenburg Johan Sigismund and his wife Anna of Prussia. Her mother had a strong position as heiress to several areas of land and as a person she was both strong-willed and energetic.
She was the daughter of John Sigismund, Elector of Brandenburg, and Anna, Duchess of Prussia, daughter of Albert Frederick, Duke of Prussia. In the year 1620, Maria Eleonora married the Swedish king Gustavus Adolphus with her mother´s consent, but against the will of her brother George William, Elector of Brandenburg, who had just succeeded her father.

Anna Maria war die älteste Tochter des Markgrafen Georg von Brandenburg-Ansbach (1484–1543) aus dessen zweiter Ehe mit Hedwig (1508–1531), Tochter Herzog Karls I. von Münsterberg. Anna Maria wurde im lutherischen Glauben erzogen. Sie heiratete am 24. Februar 1544 in Ansbach den nachmaligen Herzog Christoph von Württemberg (1515–1568).

Anna Maria was a daughter of the John George, Elector of Brandenburg (1525–1598), from his second marriage to Sabina of Brandenburg-Ansbach (1548–1575), daughter of the George, Margrave of Brandenburg-Ansbach. In 1580, she was engaged to Barnim X, Duke of Pomerania of Pomerania and on 8 January 1581, she married him in Berlin.
