Born at Shanklin, Isle of Wight, he was the son of Captain Charles Henry Hamilton, a descendant of Lt.-Gen. James Hamilton, 4th Duke of Hamilton, and Elizabeth Anne Hill.
Alfred Douglas Hamilton succeeded his kinsman, William Alexander Louis Stephen, 12th Duke of Hamilton and 9th Duke of Brandon, in 1895. He served as a Deputy Lieutenant for Lanarkshire.
