John William Polidori est né d'un père italien, Gaetano Polidori, un homme de lettres toscan, secrétaire pour un temps du dramaturge Vittorio Alfieri et d'une mère anglaise. Il a suivi des études de médecine à l'Université d’Édimbourg. Le thème de sa thèse de doctorat, qu'il présenta à l'âge de 19 ans, est le somnambulisme.
