Johann von Brandenburg-Küstrin was born 3 August 1513 in Tangermünde to Joachim I. Nestor von Brandenburg (1484-1535) and Elisabeth of Denmark (1485-1555) and died 13 January 1571 Küstrin of unspecified causes.
Johan was een jongere zoon van keurvorst Joachim I Nestor van Brandenburg en Elisabeth van Denemarken. Zijn vader had in zijn laatste wil gestipuleerd dat zijn domeinen verdeeld moesten worden tussen zijn zonen Johan en Joachim II Hector. Toen Joachim I Nestor in 1535 stierf, werd voor Johan het markgraafschap Brandenburg-Küstrin gecreëerd.
Catherine of Brunswick-Wolfenbüttel, Margravine of Brandenburg-Küstrin (1518–1574), daughter of Henry V, Duke of Brunswick-Lüneburg, wife of John, Margrave of Brandenburg-Küstrin Catherine Antonovna of Brunswick (1741–1807), daughter of Duke Anthony Ulrich of Brunswick, imprisoned by Empress Elizabeth of Russia along with her family for her entire life.
Elizabeth was the elder of two daughters of Margrave John of Brandenburg-Küstrin (1513–1571) from his marriage to Catherine of Brunswick-Wolfenbüttel (1518–1574), daughter of the Duke Henry V of Brunswick-Lüneburg. On 26 December 1558 Elisabeth married Margrave George Frederick I of Brandenburg-Ansbach-Kulmbach (1539–1603) in Küstrin.
