Gisela Agnes war das einzige überlebende Kind des Fürsten Leopold von Anhalt-Köthen (1694–1728) aus dessen erster Ehe mit Friederike Henriette (1702–1723), Tochter des Fürsten Karl Friedrich von Anhalt-Bernburg . Da ihr Vater ohne männlichen Erben geblieben war, folgte ihm Gisela Agnes’ Onkel August Ludwig als Fürst von Anhalt-Köthen.

Eleonore Wilhelmine married first on 15 February 1714 in Köthen to Prince Frederick Erdmann of Saxe-Merseburg (1691-1714), son of Christian II, Duke of Saxe-Merseburg.

Leopold was born at Dessau as the eldest son of the later Leopold II, Prince of Anhalt-Dessau, by his wife Gisela Agnes, daughter of Leopold, Prince of Anhalt-Köthen. After having lost both parents in 1751 (his mother on 20 April and his father on 16 December), the eleven-year-old Leopold inherited Anhalt-Dessau under the regency of his uncle, Prince Dietrich .
