Unlikely (although some theories suggest they are – see below). ‘Jack and Jill’ is historically a generic way of referring to a male and female pair. “Jill” was originally spelled “Gill”, which is typically a boy’s name, in the earliest version of the rhyme.
David Koechner was considered for the role of Jack before Billy Bob Thornton was cast. Melissa McCarthy was considered for the role of Jill before Amy Sedaris was cast. In their eponymous nursery rhyme, they were siblings. In the movie though, they are husband and wife.
