He was the son of Charles Calvert, 5th Baron Baltimore, the third Proprietor Governor of Maryland (1699–1751), and may have been the grandson of King George I of Great Britain.

Charles Benedict Calvert (1808-64) served as acting president of the Maryland Agricultural College in late 1859 and 1860. Calvert was founder of the Maryland Agricultural College, president of the Board of Trustees and a well-known philanthropist, planter and congressman. Enrollment increased under his brief administration, from 35 to 63 students.
