Louise Auguste Wilhelmine Amalie was born March 10, 1776 , in Hanover, where her father, Duke Charles of Mecklenburg-Strelitz, was commandant. Early in 1792, when Louise was 16, her uncle, hoping to strengthen the ties between his family and the Prussian royal family, carefully plotted a meeting between Louise and Crown Prince of Prussia.
Charles II (Charles Louis Frederick; 10 October 1741 – 6 November 1816) was ruler of the state of Mecklenburg-Strelitz from 1794 until his death. Originally ruling as duke, he was raised to the rank of grand duke in 1815. Prior to succeeding to the throne, he served as Governor of Hanover from 1776 to 1786.

Duchess Louise of Mecklenburg-Strelitz was Queen of Prussia and the wife of King Frederick William III. The couple's happy, though short-lived, marriage produced nine children, including the future monarchs Frederick William IV of Prussia and Wilhelm I, German Emperor.
Her other niece, Louise's sister, Princess Friederike of Mecklenburg-Strelitz married in 1815 Prince Ernest Augustus, Duke of Cumberland, who, in 1837, became King of Hanover, making her queen consort of Hanover. Mecklenburg-Strelitz adopted the constitution of the sister duchy in September 1755.
