Louise married Frederick Francis, Hereditary Duke of Mecklenburg-Schwerin, eldest child and only son of Duke Ludwig of Mecklenburg-Schwerin and his wife Princess Charlotte Sophie of Saxe-Coburg-Saalfeld, on 1 June 1775 in Gotha.
She was the daughter of Frederick I, Duke of Saxe-Gotha-Altenburg (1646–1691) and Magdalena Sibylle, Duchess of Saxe-Weissenfels (1648–1680). Her father was a fourth-generation descendant of John Frederick, Elector of Saxony in direct male line.

Born in 1708, Ludwig Günther II was the third of the three sons and the youngest of the thirteen children of the first Prince of Schwarzburg-Rudolstadt, Ludwig Friedrich I and Anna Sophie of Saxe-Gotha-Altenburg. He succeeded to the throne upon the death of his nephew Johann Friedrich, Prince of Schwarzburg-Rudolstadt, who had no sons.
