It was created when the Anhalt territory was divided among the sons of Prince Henry I into the principalities of Anhalt-Aschersleben, Anhalt-Bernburg and Anhalt-Zerbst in 1252. In the course of the partition, Prince Siegfried I, the youngest son of Henry I, received the lands around Köthen, Dessau, and Zerbst.

Siegfried I comes from the Ascanian family and was the third son of Henry I, Prince of Anhalt, and Irmingard of Thuringia. He ruled from 1252 to 1298.
The territory belonging to this branch of the family had been divided in 1396, and after the acquisition of Bernburg Prince George I made a further partition of Zerbst (Zerbst and Dessau).
