Gwyn had two sons by King Charles: Charles Beauclerk (1670–1726) and James Beauclerk (1671–1680). Charles Beauclerk was created Earl of Burford and Duke of St. Albans; Murray de Vere Beauclerk, 14th Duke of St. Albans is her descendant, and the current holder of the duchy .

Her childhood occupations have been variously described as bawdyhouse servant, a street hawker of herring, oysters, or turnips, and cinder-girl. Her mother was drowned in a pond at Chelsea, probably while drunk, in July 1679.
Nell settled her mother in a house in Chelsea, where, in July 1679, overcome by brandy, Mrs. Gwyn fell into a nearby brook and was drowned. Of all the mistresses of Charles II, Nell was the only one beloved by the public.

She was soon a popular favourite in light comedy, starring opposite the actor Charles Hart, who was her lover, and scoring a triumph in Dryden’s Secret Love in 1667.
