Gladstone was the fourth son of Sir John Gladstone, 1st Baronet (1764-1851) and was born in Liverpool, though his family had strong Scottish ties. The Gladstones were a rich family, with a fortune built on the corn and tobacco trade and West Indian sugar plantations which employed some 2,500 enslaved Africans in 1833.

Born at Leith on the 11 th of Dec. 1764, he commenced business there at an early age, but soon removed to Liverpool, where he amassed considerable riches by his enterprise, industry, and skill, and was munificent in their disposal
Sir John Gladstone, 1st Baronet, of Fasque and Balfour in the County of Kincardine, died at Fasque House in December 1851, 4 days shy of his 87th birthday, and was buried at St Andrew's Episcopal Church at Fasque. He was succeeded by his eldest son, Sir Thomas Gladstone, 2nd Baronet.

The elder brother of the Liberal Prime Minister William Ewart Gladstone, he was born in Liverpool, the eldest son (and second child) of the wealthy Scottish businessman Sir John Gladstone, 1st Baronet and his second wife Anne MacKenzie Robertson. He was educated at Eton College from 1817. Tom hated Eton, its discipline harsh and irreligious.
