Nissl worked with Alzheimer in Frankfurt, and they both moved to Munich in order to advance their researches on brain pathology. He also worked with Brodmann, whose cytoarchitecture was based largely on his studies using the Nissl stain. The portrait of Nissl is combined with brain tissue stained by his method.

Franz Nissl (9 septembre 1860 à Frankenthal - 11 août 1919 Munich) est un neurologue neuropathologiste et psychiatre allemand. Il donne son nom à une technique de coloration particulièrement utile en neuropathologie car elle ne fait apparaître que les corps cellulaires des neurones ( péricaryon ) et non leurs prolongements ( axones ).
Nissl y Alzheimer se embarcaron juntos en una extensiva investigación de la patología del sistema nervioso, estudiando en particular la anatomía patológica y normal de la corteza cerebral, trabajo que dio origen a un tratado de seis volúmenes denominado "Estudios Histológicos e Histopatológicos de la Corteza Cerebral", publicado entre 1906 y 1918.

Il metodo Nissl impiega anilina basica per colorare l'RNA di blu, ed è utilizzata per evidenziare caratteristiche strutturali dei neuroni. La sostanza di Nissl ( reticolo endoplasmatico rugoso ) appare blu scuro in quanto maggiormente ricca di RNA, mentre il citoplasma appare di colorazione marmorizzata.
