Joker was done for all intents and purposes by the end of The Dark Knight. And as stated previous, Nolan didn't imagine him in the third film anyway. That being said, if Ledger was still alive, he could have potentially replaced one or two of the many characters introduced to the film, maybe have it tied a bit more firmly to The Dark Knight than it ended up being.
