Born in Weimar, Dorothea Maria was the youngest daughter of Duke William IV of Saxe-Weimar from his marriage to Eleonore Dorothea of Anhalt-Dessau, the daughter of Prince John George I of Anhalt-Dessau. She was named after her maternal grandmother Dorothea Maria of Anhalt, who had already died at that point in time.

She first pursued a clerical carreer and was from 1568 until 1593 abbess of Gernrode and Frose. She married Johann von Sachsen-Weimar in 1593 and bore him eleven children.

She was the last abbess of Gernrode (r.1593-1614). After leaving the abbey, Sophie Elisabeth married George Rudolf of Liegnitz and was duchess of Liegnitz until her death in 1620.
