Louise was the daughter of Margrave Frederick Henry, Margrave of Brandenburg-Schwedt and his wife, Princess Leopoldine Marie of Anhalt-Dessau. By birth, Louise Henriette belonged to Brandenburg-Schwedt line of the House of Hohenzollern. Biography. She was educated in Prussia, together with her sister, Friederike Charlotte of Brandenburg-Schwedt.

Hon föddes prinsessa av Brandenburg och var den äldsta dottern till kurfurst Georg Vilhelm av Brandenburg och Elisabeth Charlotta av Pfalz. Louise Charlotte gifte sig 6 oktober 1645 med hertig Jakob av Kurland.
Louise Charlotte var opptatt av politikk, og korresponderte flittig med kolleger i utlandet. Forhandlingene mellom Brandenburg, Polen, Russland og Sverige fant sted i Mitau i hennes regjeringstid. Hertugparet og barna ble i årene 1658–60 holdt som fanger av svenskene først i Riga og senere i Ivangorod .
