Cölln nevnes første gang 28. oktober 1237, mens Berlin ble nevnt første gang i 1244. Byene ble slått sammen i 1707. I middelalderen hadde Cölln 1 400 innbyggere. Som bydel i Berlin hadde Cölln i 1871 16 554 innbyggere. I 1910 var innbyggertallet sunket til 1 400 og i 1920 ble Cölln innlemmet i den daværende bydelen Mitte.
