Laura fue la primogénita de Lorenzo Mancini y Girolama Mazzarini, hermana del cardenal Mazarino. Gracias a la influencia de su tío, consejero de Luis XIV, Laura y su familia fueron invitados a Francia, abandonando Roma en 1653, tras la muerte de su padre, junto con sus primas Laura y Ana María .

Eran hijas de las dos hermanas del cardenal, Laura Margherita Mazzarini (fallecida en 1685), esposa de Girolamo Martinozzi, y Girolama Mazzarini (1614-1656), esposa de Michele Lorenzo Mancini. 2 En 1647, el cardenal Mazarino trajo a Laura Margherita y sus dos hijas, Anne Marie y Laura, de Italia a París.
Su madre, Girolama Mazzarini, era una de las dos queridas hermanas del poderosísimo valido de Ana de Austria: el Cardenal Jules de Mazarin (aunque ha pasado a la historia con esta versión afrancesada de su nombre, en realidad había nacido Giulio Raimondo Mazzarini, en un tranquilo pueblo de los Abruzzos italianos).
