Honora was born about 1675 at Portumna Castle, County Galway. She was the youngest child of William Burke and his second wife, Helen MacCarty. Her father was William Burke, 7th Earl of Clanricarde. The Burkes (originally De Burgh) were an Old English family long-established in Connacht.

She married Ulick Bourke, 3rd Earl of Clanricarde, son of Richard Bourke, 2nd Earl of Clanricarde and Margaret O'Brien, on 25 November 1564 at Anthenry, County Galway, Ireland. Children of Honora Burke and Ulick Bourke, 3rd Earl of Clanricarde.

Richard Burke, IV conde de Clanricarde (también Richard de Burgh) (1572 – 12 de noviembre de 1635) fue un noble y político irlandés. Era hijo de Ulick Burke, III conde de Clanricarde y Honora Burke.
Clanricarde, as he now was, married Honora Burke, daughter of John Burke of Clogheroka, on 25 November 1564 at Athenry, County Galway. Ulick and Honora had six sons: Richard (died an infant) [2] Richard Burke, 4th Earl of Clanricarde (1572–1635) Thomas, who married Ursula Malby, daughter of Sir Nicholas Malby and widow of Anthony Brabazon.
