Nacida en Mantua en 1598, Leonor era la hija menor del duque de Mantua Vicente Gonzaga y Leonor de Médicis . Leonor pasó la infancia en la corte de Mantua. Famosa por su singular belleza, se casó con Fernando en Innsbruck el 2 de febrero de 1622. La anterior esposa de Fernando, María Ana de Baviera, había muerto el 8 de marzo de 1616.

During this time, acting on behalf of her brother, she began secret negotiations for the marriage of her nephew Francesco Gonzaga, Hereditary Prince of Mantua and Princess Margaret of Savoy, the eldest daughter of Charles Emmanuel I, Duke of Savoy; the marriage was concluded in 1608. At the end of 1601, Margarita returned to Mantua from Montferrat.
