A member of the House of Guise founded by Claude, Duke of Guise, he was a Prince of Lorraine as a male line descendant of René II, Duke of Lorraine. In his youth he was not expected to succeed to the Duchy-Peerage of Elbeuf as his father had a son (another Charles, 1650–1690) from a previous marriage as well as his oldest full brother Henri Frédéric.

René II de Lorraine-Guise, né à Joinville le 14 août 1536, mort le 14 décembre 1566, est baron d'Elbeuf de 1550 à 1554, puis marquis d'Elbeuf de 1554 à 1566. Il est fils de Claude Ier, duc de Guise, d'Aumale, baron de Mayenne et d'Elbeuf et sire de Joinville, et d'Antoinette de Bourbon-Vendôme.
