La universidad fue creada en 1817 mediante la fusión de la Universidad de Wittenberg (fundada en 1502) y la Universidad de Halle (fundada en 1694 ). La universidad debe su nombre al reformador protestante Martín Lutero, quien fue profesor en Wittenberg.
