Sophia Charlotte was born on 19 May 1744. She was the youngest daughter of Duke Charles Louis Frederick of Mecklenburg, Prince of Mirow (1708–1752), and his wife Princess Elisabeth Albertine of Saxe-Hildburghausen (1713–1761). Mecklenburg-Strelitz was a small north-German duchy in the Holy Roman Empire.

Princess Sophie Charlotte was born in 1744 in the Palace of Mirow in a little known principality of Mecklenberg-Strelitz, in what is now part of Germany. It was an idyllic location in which to grow up. The castle, on a lake and surrounded by woods, resembled something out of Beauty and the Beast.
