Annie Potts se graduó en Bellas Artes del Colegio Stephens y en 1978 protagonizó, junto a Mark Hamill, Corvette Summer, por la cual obtuvo una nominación al Globo de oro a Mejor actuación debut femenina. Logró reconocimiento cuando interpretó el papel de Janine Melnitz en las películas Los cazafantasmas (1984) y Los cazafantasmas 2 (1989).
