Nacido en Londres el 7 de septiembre de 1795, Polidori siguió los pasos de su abuelo y a los dieciséis años empezó sus estudios de medicina en la Universidad de Edimburgo. A los diecinueve obtuvo el titulo tras presentar un tesis sobre el sonambulismo.
