William Frederick was the second son of Ernest Casimir I, Count of Nassau-Dietz and Sophia Hedwig of Brunswick-Lüneburg. He married Countess Albertine Agnes of Nassau, the fifth daughter of Frederick Henry, Prince of Orange on 2 May 1652 in Cleves. They had three children:

Henriette Louise (20 June 1623 – 24 August 1650) married on 21 August 1642 with Margrave Albert II of Brandenburg-Ansbach Leopold Frederick (30 May 1624 – 15 June 1662), his successor After Elisabeth Magdalena's death, Louis Frederick married on 15 May 1625 with Anna Eleonora, daughter of Count John Casimir of Nassau-Weilburg (1677-1702).
His godparents were his father's cousin and cousin-in-law, the Duke and Duchess of Saxe-Gotha-Altenburg; and the Duke of Gloucester's second cousin, the Margrave of Brandenburg-Ansbach. [2] During his stay in Stockholm in 1802–1803, William's interest and rumoured affair with Aurora Wilhelmina Koskull attracted a lot of attention, and he reportedly had plans to marry her.
His first marriage in 1570 was to Catherine, Princess of Brandenburg-Küstrin (1549–1602), daughter of John I, Margrave of Brandenburg-Küstrin, and Catherine, Duchess of Brunswick-Wolfenbüttel. Joachim Frederick's second marriage, in 1603, was to Eleonore of Hohenzollern (1583–1607), daughter of Albert Frederick, Duke of Prussia and Mary Eleonore of Cleves.
