02/03/2022 · It was designed to evaluate the efficacy and safety of guanfacine (1, 2, 3, and 4 mg/day) when co-administered with long-acting psychostimulants (amphetamine, lisdexamfetamine, methylphenidate, dexmethylphenidate) in children and adolescents aged 6-17 years with a diagnosis of ADHD and a suboptimal, partial response to psychostimulants.
Initially 1 mg once daily; adjusted in steps of 1 mg every week if necessary and if tolerated; maintenance 0.05–0.12 mg/kg once daily (max. per dose 4 mg), for optimal weight-adjusted dose titrations, consult product literature. For Child 13–17 years (body-weight 41.5–49.4 kg)
An initial phase 1 randomized, open-labeled, single-dose, crossover pharmacokinetic study was done in 52 healthy adults, ages 18–55. GXR half-life is 16.6–17.5 hours. The pharmacokinetics of GXR is generally linear for dosage from 2 to 4 mg, but there is a less than 2-fold increase between the 1–2 mg doses.
